My sister has a Fido account with two numbers associated with it. It is possible to port only one number without Speakout cancelling the whole Fido account, right?
Do I activate the new Speakout sim before porting? What info does Speakout need for the porting process? Only the Fido account + Fido number?

You activate the Speakout SIM first, and Speakout will assign a number to you. You can then get Speakout to port in your FIDO number.
Normally, I would not recommend you talk to Fido, but in this case, you are better off having them know what you are doing. I doubt Fido would cancel the whole account, but who knows what they could do. Speakout really has nothing to do with cancelling your Fido account, they are only taking the number. What Fido does with the Fido account is another story, and you should talk to Fido to get clarity on the matter.

If you are porting a number to Speakout, your Speakout balance would remain intact. The only difference is you will get a new ported in number, and lose the old number Speakout previously assigned to you.
If you were going to port out your Speakout number to another provider, that would cancel the Speakout account. The Speakout SIM would not longer be active, and any balance that was previously on the Speakout account would be no more.
I do not know if you can transfer a balance from one Speakout SIM to another. I would probably talk to a Speakout CSR to get clarity.

number was successfully ported. It only lists the old Speakout number (error message) with no balance on the website! If I add the Fido number back in, would the balance come back since it already ported onto the Speakout sim? I have to reverify it?
[edit]Never mind. had to manually enter the number and reverify on the website. Balance is still there. Phew! I thought the reps would've add the ported number automatically to your account.
Thanks again for your help Bridonca!
